A Review of Various Manufacturing Approaches for Developing Amorphous Solid Dispersions

In recent years amorphous solid dispersions (ASD) has gained a tremendous response for improving the solubility of poorly water-soluble drug substances. Despite stability challenges, various ASD commercial products have been successfully launched into market over last two decades. Among manufacturing approaches, hot melt extrusion (HME) and spray drying techniques attracted industries attributing to their simple processes. addition, KinetisolÒ, solvent-free approach, is also being most widely investigated developing ASDs since thermal exposure time formulations significantly less compared with process. KinetisolÒ can be employed thermolabile Another solvent-based technique, electrospinning, nanofibers-based ASD. However, much research warranted electrospinning process before implementing it in manufacturing. Various critical factors such as drug-polymer solubility, polymer, interactions, type process, storage conditions need considered stable robust formulation. This review mainly focuses on advanced technologies ASDs, namely HME, drying, along note that affect formulations.
